Record winners Bayern Munich will face third-league side Saarbrücken, while Borussia Dortmund will also face Hoffenheim in the Bundesliga.
League leaders Bayer Leverkusen, meanwhile, will travel to third-tier Sandhausen.
The other all-Bundesliga match is VfB Stuttgart-Union Berlin.
The second round matches will be played on October 31st and November 1st.
